Beraprost pojačava proizvodnju antigen-specifičnih izotipa IgG bez moduliranja stvaranja zametnih stanica B stanica i sazrijevanja afiniteta.

Beraprost enhances production of antigen-specific IgG isotypes without modulating germinal center B cell generation and the affinity maturation.
Priznaju se sposobnosti imunološke regulacije lipidnih medijatora. Prostaglandini (PG) su izvedeni iz fosfolipida ciklooksigenazom-2 (COX-2) u upalnim tkivima. Prvenstveno na temelju naših ranijih saznanja koja upućuju na imuno -modulirajuću vježbu prostaciklina, pretpostavili smo da PG -ovi promiču humoralne imunološke odgovore in vivo.
Kako bismo provjerili ovo nagađanje, ispitali smo utjecaj analoga prostaciklina analoga beraprosta i PGE2 na odgovore protutijela izazvane imunizacijom miševa hemocijaninom u ključaonici (KLH). Beraprost je korišten kao rezultat izuzetno nestabilnog svojstva prostaciklina . Naši rezultati potvrdili su da je beraprost zasigurno promicao proizvodnju anti-KLH antitijela, koja je ovisila o dozi, a posebno na beraprost kao rezultat PGE2 nije modulirala odgovor antitijela za razliku od kontrola.
Pojačani utjecaj beraprosta reproduciran je unutar sekundarnih odgovora, što upućuje na to da je beraprost na cijelu glavnu reakciju značajno utjecao na tehnologiju reminiscencije B stanica. Evaluacija izotipova anti-KLH antitijela otkrila je da je beraprost potaknuo proizvodnju subzotipova IgA i IgG, ali ne i IgM.
Ipak, primjena beraprosta nije utjecala na tehnologiju zametnih srednjih B stanica i afinitet anti-KLH antitijela. Da bismo provjerili ove ishode, imunizirali smo COX-2 nokaut miševe s KLH-om i analizirali jesu li rezultati s miševima divljeg tipa preslikani u odsutnosti PG-a.
Prvi i sekundarni odgovor protutijela znatno su oslabljeni kod KO životinja. Stupnjevi pod-podtipova i IgA anti-KLH IgG značajno su smanjeni kod KO miševa, dok su ti IgM slični kontroli. Ovi ishodi otkrivaju neprepoznatu izvedbu PG -a unutar humoralnih imunoloških odgovora.
Ekspresija specifična za tip i izotip i regulacija β-tubulina u glavnim stanicama koje ulaze u njuh i Schwannovim stanicama in vitro.
Stanice koje ulaze u miris (OEC) i Schwannove stanice (SC) blisko su povezane stanice sa svojstvima koja potiču regeneraciju. Usporedna evaluacija ekspresije gena posebno je povezana jer bi mogla razjasniti rezultate i informacije specifične za stanični tip pomoću svake vrste stanica u određene medicinske funkcije.
Unutar trenutnog ispitivanja ciljali smo na ekspresiju izotipa β-tubulina u velikim odraslim psećim glijama kao translacijski divovski životinjski maneken. β-tubulini do sada su proučavani prvenstveno u tumorima koji nisu neuroni i implicirani u tumorigenom razvoju. Ovdje predstavljamo da su glavni OEC i SC eksprimirali mRNA izotipa βII-V izotipa.
Zanimljivo je da je ekspresija mRNA i proteina βIII-tubulina bila prekomjerna u OEC-ima, a niska u SC-ima, dok je faktor razvoja fibroblasta-2 (FGF-2) inducirao njegovu smanjenu regulaciju u svakoj vrsti stanica u istoj mjeri. To se razlikovalo od mRNA βV-tubulina koja je bila podjednako izražena u svakoj vrsti stanica i nepromijenjena FGF-2.
Imunocitokemijska procjena otkrila je da OEC kulture sadrže bolji udio βIII-tubulin-pozitivnih stanica u usporedbi sa SC kulturama. Dodavanje FGF-2 smanjilo je raznolikost stanica pozitivnih na βIII-tubulin u svakoj kulturi i značajno povećalo udio stanica s multipolarnom morfologijom.
Uzeti zajedno, prikazujemo ekspresiju specifičnu za stanični tip (βIII) i izotip specifičnu regulaciju (βIII, βV) izotipova β- tubulina u OEC-ima i SC-ima. Dok diferencijalna ekspresija βIII-tubulina u glavnim vrstama glija stanica s ekvivalentnim proliferativnim ponašanjem zalaže se za nove sposobnosti koje nisu povezane s tumorskim razvojem, robusna ekspresija βIII-tubulina u OEC-ovima mogla bi pomoći razjasniti posebna svojstva ove vrste glijalnih stanica.

CD8aa i -αβ  izotipovi  jednako su regrutirani na imunološki sinapse preko njihove sposobnosti da se vežu na MHC klase I.

Komplementacija bimolekularne fluorescencije korištena je za konstruiranje molekula CD8 kako bi se dimeri CD8αα i CD8αβ mogli neovisno vizualizirati na podu T stanice tijekom prepoznavanja antigena. Koristeći ovu metodu, pokazujemo da se CD8αα regrutira u imunološku sinapsu gotovo pored CD8αβ, međutim, kao rezultat kinaze Lck asocira prvenstveno na CD8αβ u lipidnim splavovima, CD8αα je slabiji ko-receptor.
Tijekom prepoznavanja robusnog CD8αα liganda H2-TL, CD8αα se prvenstveno regrutira. Dakle, regrutiranje 2 vrste CD8 korelira s njihovim relativnim vezanjem za pristupačne ligande, više nego s mogućnostima ko-receptora vrste CD8.

Ekspresiju Tubb3, izotipa beta-tubulina  , reguliraju androgeni u stanicama Sertolija miša i štakora.

Naša ranija evaluacija miševa nokautiranih stanica SCTO (SCARKO) iz Sertolijevih stanica otkrila je da su brojni citoskeletni elementi mogući cilj kretanja androgena. Ovdje smo otkrili da je jedan od ovih elemenata, izotip beta-tubulina Tubb3, različito reguliran u testisima od SCARKO miševa (u odnosu na kontrole s leglom) od postnatalnog 10. dana do zrelosti.
Gen Tubb3 je u tom pogledu ekskluzivan, budući da 10. dana AR ne značajno regulira različite gene beta-tubulina. Dodatno smo okarakterizirali androgenu regulaciju Tubb3 in vivo i in vitro i pokazali da je to očuvana karakteristika kod svakog miša i štakora.
Kako bismo analizirali jesu li androgeni odmah regulirali ekspresiju Tubb3, pregledali smo dijelove odgovora na androgene (AREs) unutar gena Tubb3. In silico evaluacija otkrila je prisutnost 4 ARE motiva u Tubb3 intronu 1, od kojih se dva vežu za AR in vitro.
Mutacija jedne od ovih (ARE1) snažno je smanjila ekspresiju reporterskog gena ovisnu o androgenima. Ovi ishodi, zajedno s otkrićem da se AR veže za područje ARE Tubb3 in vivo, govore da je Tubb3 izravni cilj AR. Naše znanje jača konkurenciju da androgeni ostvaruju svoje rezultate na spermatogenezi, djelomično, modulacijom citoskeleta Sertolijevih stanica.
Androgenska regulacija beta-tubulina dodatno je opisana u neuronima, učvršćujući već prepoznatu sličnost u skupini mikrotubula u procesima i neuronima Sertolijevih stanica, jednoj različitoj vrsti stanica tijekom koje se smatra da se eksprimira Tubb3.Izotoglobulinski izotipi  holštajn krava u laktaciji kategorizirani su kao prekomjerni uobičajeni i niski imunološki odgovori tipa 1 ili -2.
Zarazne bolesti štetne su za dobrobit i ekonomski sustav stočarske djelatnosti. Promatranja dokaza goveda protiv čistih infekcija implicirala su izvedivost uzgoja stoke za otpornost na bolesti. Istraživanja svinja odabranih za imunološke odgovore posredovane protutijelima (AMIR) i stanicama (CMIR) pokazala su povišenu imunološku reakciju koja sugerira povećanu sigurnost za svaki odgovor vrste 2, odnosno sorte 1.
Štoviše, čiste ili sintetičke infekcije goveda smatraju da je proizvodnja specifičnih izotipova imunoglobulina (Ig) M, IgG1 i IgG2 bitna za zaštitu od patogena. U stvarnosti, omjeri IgG1/IgG2 ponekad se koriste za određivanje prevladavaju li odgovori 1 (CMIR) ili 2 (AMIR) nakon imunizacije ili infekcije.
Ciljevi ovog ispitivanja su posljedica ove činjenice; (1) procijeniti reakcije pristranosti izotipa Ig izolata na Candida albicans i lizozim bjelanjka kokošijeg jaja (HEWL) u krava kategoriziranih kao prekomjerno odgovorne osobe (HR), uobičajene osobe s odgovorima (AR) ili niske reakcije (LR) prvenstveno na temelju AMIR ili CMIR; (2) kako bi se utvrdilo jesu li ocjene prvenstveno temeljene na IFN-γ (citokin vrste 1) i DTH odgovorima analogne po ocjeni; i (3) za procjenu IFN-γ, Ig izotipova i DTH korelacija.
Otkriveni su odgovori protutijela na HEWL i DTH na C. albicans tako da su krave fenotipski kategorizirane kao HR, AR i LR za AMIR ili CMIR s vitalnim varijacijama (p ≤ 0,05) među kategoriziranim timovima. IFN-γ izazvan C. albicans dopušta klasifikaciju krava, od kojih su neke imale istu ocjenu kao i DTH odgovor. Donji omjer IgG1/IgG2 bio je prema pročišćenom antigenu C. albicans (candin), međutim nisu primijećene varijacije u omjerima anti-HEWL ili anti-kandin IgG1/IgG2 između kategoriziranih timova.
